Caribou brings a distinct, defensible technical bet to CRISPR — its chRDNA hybrid RNA-DNA guides, which replace part of the RNA guide with DNA to improve editing specificity and enable the high-efficiency multiplex edits and insertions that off-the-shelf cell therapies require — and it has now focused that platform on its two most clinically-de-risked assets. In April 2025 Caribou made the hard, disciplined choice to prioritize: it discontinued its lupus and AML programs plus preclinical research, cut roughly 32% of staff, and concentrated capital on lead allogeneic CAR-T programs vispa-cel (relapsed/refractory B-cell non-Hodgkin lymphoma) and CB-011 (relapsed/refractory multiple myeloma), extending runway into the second half of 2027. The clinical signal supports the focus: vispa-cel's ANTLER Phase 1 data in second-line large-B-cell lymphoma showed efficacy and durability the company characterizes as on par with autologous CAR-T — the holy grail for an off-the-shelf therapy that promises speed, scalability, and access an autologous product cannot — and CB-011 posted a 92% overall response rate and 91% MRD-negative rate in early multiple-myeloma data. Vispa-cel carries FDA RMAT, Fast Track, and Orphan Drug designations. Co-founded by Nobel laureate Jennifer Doudna and led by Rachel Haurwitz, and public since 2021 with a strong cash position, Caribou has a differentiated editing chemistry, focused late-ish-stage assets, and runway to reach value-inflecting data.

The 32% workforce cut and pipeline gutting in April 2025 is the bear case in miniature: Caribou was forced to abandon multiple programs and shrink to survive, a defensive retrenchment that signals capital pressure and narrows the company to binary outcomes on two assets. Allogeneic CAR-T is the harder path in cell therapy — off-the-shelf products have historically struggled with durability and persistence versus autologous CAR-T because the edited donor cells face immune rejection, and "on par with autologous" from early Phase 1 data must survive larger, longer trials before it means anything. Both indications are ferociously competitive: B-cell lymphoma and multiple myeloma already have multiple approved autologous CAR-T therapies (from Gilead/Kite, BMS, J&J/Legend, Novartis) plus other allogeneic challengers, so Caribou must prove not just efficacy but that its off-the-shelf convenience justifies switching in a crowded market where autologous products are entrenched and reimbursed. Runway extending only into 2H 2027 means Caribou will need to raise again — as a small-cap gene-editing stock in a depressed sector — before either program reaches approval, and the chRDNA specificity advantage, while scientifically elegant, has not yet demonstrated a decisive clinical or commercial edge over conventional editing. A company that just cut a third of its staff and bet everything on two allogeneic assets in saturated indications carries acute clinical and financing risk.
