• Metagenomics-Derived Editing Toolbox

  Metagenomi's core platform mines vast metagenomic sequence datasets to discover new gene-editing proteins, yielding a portfolio that includes CRISPR nucleases, ultra-small base editors, and large-payload integration systems. The toolbox is designed so that the right tool can be matched to each genomic target — from single-base correction to inserting entire genes — rather than forcing every disease onto one enzyme. Partners license these systems for their own pipelines; Ionis has advanced collaboration programs into lead optimization using Metagenomi editors.

• RIGS and CAST Systems

  RNA-mediated integration systems (RIGS) and CRISPR-associated transposase (CAST) systems are Metagenomi's tools for large-cargo gene insertion — writing entire functional genes into the genome at a chosen site rather than only correcting individual bases. This addresses diseases caused by many different mutations in a single gene, where one whole-gene insertion can substitute for hundreds of distinct corrections. In May 2024 Metagenomi regained full development rights to its wholly owned base-editing and RIGS systems after concluding its Moderna collaboration.
